چکیده

The aim of the study was to observe changes in the diversity of bacterial community in maize rhizosphere influenced by organic and mineral fertilization. Four variants of fertilization were tested vermicompost (VC) at recommended annual dose 40t*ha, doubled annual dose of VC, recommended dose of ammonium saltpeter with dolomite (LAD 27) and combination of VC and LAD 27. Experiment was conducted with potted maize plants in controlled conditions of greenhouse during 74 days. Using PCR-DGGE method, we investigated differences in total bacteria community as well as in community of ammonia oxidizing bacteria. Based on occurrence of operative taxonomic units (OTU) we found differences in bacterial species spectra among fertilization variants. The highest Shannon ́s biodiversity index was observed in variant with VC addition in dose 80 t*ha.The fertilizers effect on diversity of ammonia oxidizing bacteria was not significant however in each variant with vermicompost addition was the occurrence of new specific OTU observed. This OTU was identified as Nitrosospira sp. It was proven that some bacterial species introduced to soil with vermicompost addition can survive for at least 74 days and these bacteria can influence basic functions of soil microbiocenosis in nitrogen cycle.

Comparative analysis of bacterial community structure in the rhizosphere of maize by high-throughput pyrosequencing

In this study, we designed a microcosm experiment to explore the composition of the bacterial community in the rhizosphere of maize and bulk soil by sequencing the V3-V4 region of the 16S rRNA gene on the Illumina system. 978-1239 OTUs (cut off level of 3%) were found in rhizosphere and bulk soil samples. Rhizosphere shared features with the bulk soil, such as predominance of Acidobacteria, Pro...

Cultivation Effect of Chitinase-Transgenic Cotton on Functional Bacteria and Fungi in Rhizosphere and Bulk Soil

Background: In consideration for the increasing widespread use of genetically modified (GM) crops, one of the important issues for assessment is the effect of GM crops on soil microbial communities Objectives: In this study, T2 chitinase-transgenic cotton (line #57) and its non-transgenic line were investigated for bacterial and fungal dynamics...

Multitrophic Interaction in the Rhizosphere of Maize: Root Feeding of Western Corn Rootworm Larvae Alters the Microbial Community Composition

BACKGROUND Larvae of the Western Corn Rootworm (WCR) feeding on maize roots cause heavy economical losses in the US and in Europe. New or adapted pest management strategies urgently require a better understanding of the multitrophic interaction in the rhizosphere. This study aimed to investigate the effect of WCR root feeding on the microbial communities colonizing the maize rhizosphere. METH...
